Early effects of glucocorticoid treatment on amino acid metabolism in lymphosarcoma P1798.

چکیده
Administration of 9a-fluoroprednisolone causes an early (3 hr) decrease of basic amino acid incorporation into CS-P1798 lymphosarcoma protein. Decreased utilization of neutral amino acids is observed after 6 hr. These effects are accompanied by expansion of the intratumoral free amino acid pool in CS-but not in CR-P1798. Thus, decreased incorporation of radioactive amino acids in CS-P1798 appears to result from pool dilution of the tracers. It is suggested that the increase in intratumoral levels of free amino acids is a consequence of steroid-induced breakdown of tumor protein and that this effect may be related to tumor regression.

متن کاملInhibition of thymidine and basic amino acid metabolism in P1798 lymphosarcoma by L-asparaginase.
In vivo metabolism of lysine and arginine is markedly inhibited (p < 0.025) in P1798 lymphosarcoma within 1 hr after intraperitoneal injection of 1 unit of Escherichia coli L-asparaginase II. Two hr after treatment, there is substan tial inhibition of thymidine transport but no effect on incorporation of thymidine. None of these effects were observed in the L-asparaginase-resistant subline of t...
متن کاملEffects of cortisol on DNA metabolism in the sensitive and resistant lines of mouse lymphoma P1798.
Administration of cortisol to mice bearing the cortisol sensitive or -resistant lines of lymphosarcoma P1 798 resulted in a marked inhibition of incorporation of labeled deoxy nucleoside precursors into tumor DNA only in the sensitive tumor. This effect is one of the earliest biochemical changes see n in lymphosarcoma P1798 following glucocorticoid treatment. It is dependent on both time and do...
متن کاملCell membrane sialoglycopeptides of corticoid-sensitive and -resistant lymphosarcoma P1798.
Cells of corticoid-sensitive (CS) and corticoid-resistant (CR) lymphosarcoma P1798 were labeled in vivo with either [14C]- or [3H]fucose before and after treatment with 9-alpha-fluoroprednisolone (9-FP). Labeled glycopeptides, derived from isolated, Pronase-digested cell membranes of both tumors were separated by gel filtration on Bio-Gel P-10 by a double-label technique. Elution profiles of CS...
متن کاملNuclear Binding of Steroid-Receptor Complex to Lymphosarcoma P1798 Resistant and Sensitive Cells and Effect of Concanavalin A on Receptor Levels1
Glucocorticoid-resistant P1798 cell lines have been found to contain levels of glucocorticoid receptor comparable to receptor levels in glucocorticoid-sensitive P1798 cells. Pre viously, most of the P1798 resistant cells examined were found to contain low levels of glucocorticoid receptor, and this was thought to account for the resistance of these cells to glucocorticoid treatment. Resistant c...
